Probability-based approaches to witnessing exciton delocalization in microtubules can be misleading, particularly when the microtubule is an open system, because the simultaneous presence of an exciton at two different tryptophan (Trp) sites with nonzero probabilities does not necessarily imply delocalization if the off-diagonal term of the density matrix coupling those sites is zero.
